چکیده

Gentamicin C complex, produced by Micromonospora purpurea1*2), is composed of three components, Cla, C2 and Q (Fig. 1). These components are methylaminoglycosides, which are distinct from other related antibiotics such as the kanamycins, neomycinsand paromomycins. As seen in Fig. 1, gentamicins Cla, C2, and Q possess one C-methyl and one N-methyl group, two C-methyl and one N-methyl groups and two C-methyl and two N-methyl groups, respectively. Presuming that L-methionine is the donor of methyl groups of gentamicins, it may be assumed that most of the radioactivity from L-methionine-methyl-14C would be incorporated into the Q component and least into the Cla component. Experimentally, incorporation of L-methionine-methyl14C into the gentamicins was found to be highest as compared to other radioactive presumed

[Methyl-14C]-choline incorporation into nude mice bearing tumor xenografts-correlation with [methyl-3H]-thymidine: A pilot study

Background: Many in vitro studies suggested that choline incorporation into many types of tumors is related to cell proliferation. Whether in vivo choline incorporation is related also to cell proliferation or not was my question. Therefore, the aim of this pilot study was to investigate the relationship of in vivo [methyl- 14C]-choline incorporation and in vivo [methyl-3H]- thymidine incorpora...

Incorporation of L-methionine-methyl-14C into gentamicins. II. Large-scale preparation of methyl-14C-gentamicins.

Methyl-14C-gentamicins were prepared from incubation of Micromonospora purpurea in the presence of added methyl-'4C-L-methionine, and products with high specific radioactivity were isolated in satisfactory yields. In the previous paper1), we have shown in shake flask fermentation a high and selective incorporation of radioactivity from methyl-14C-L-methionine into gentamicins. Consequently, we ...

Synthesis of carbon-14 and tritium labeled sagamicin.

Sagamicin, an aminoglycoside antibiotic, was labeled with tritium by means of a platinum catalyzed hydrogen exchange reaction and by carbon-14 in two biosynthetic procedures. 14C-Methyl-L-methionine was used as the radioactive precursor in these biological processes involving Micromonospora sagamiensis. The distribution of radioactivity in 14C-sagamicin and gentamicin C1 was studied by mild aci...

Mevinolinic acid biosynthesis by Aspergillus terreus and its relationship to fatty acid biosynthesis.

Mevinolinic acid, the open acid form of mevinolin, which is a metabolite of Aspergillus terreus, has been shown to be a competitive inhibitor of the enzyme 3-hydroxy-3-methylglutaryl coenzyme A reductase (Alberts et al., Proc. Natl. Acad. Sci. U.S.A. 77:3957-3961, 1980). The biosynthesis of mevinolinic acid was studied by examining the incorporation of [1-14C]acetate and [methyl-14C]methionine ...

Comparative metabolism of L-methionine, DL-methionine and DL-2-hydroxy 4-methylthiobutanoic acid by broiler chicks.

1. Metabolism, in broiler chicks, of DL-2-hydroxy 4-methylthiobutanoic acid (DL-HMB), DL-methionine and L-methionine was compared in vivo using 14C-labelled tracers. 2. The distribution of L-[1-14C]methionine and DL-[1-14C]HMB in the major body tissues was examined for a period of 120 min after administration. 3. The relative oxidation (14CO2 exhaled), excretion and incorporation into tissue pr...
